I. Background

     激活素 A（Activin A）是轉(zhuǎn)化生長(zhǎng)因子-β（TGF-β）超家族的重要成員，是一種多能因 子，參與過敏、自身免疫性疾病、癌癥等免疫失調(diào)疾病的發(fā)生。 激活素 A 通過兩種 I 型受體和兩種 II 型受體發(fā)出信號(hào)，當(dāng)它們與配體結(jié)合時(shí)，組裝成 最終的受體復(fù)合物。I 型受體包括 1A 型激活素受體（或激活素受體樣激酶 2，ALK2）、 1 B 型激活素受體（或 ALK4）和 1C 型激活素受體（或 ALK7）。Activin-A 有利于 ALK4 結(jié) 合，而對(duì) ALK2 和 ALK7 的親和力較低。II 型受體是 Activin 受體 IIA 型（ActRIIA）和 Act ivin 受體 IIB 型（ActRIIB），其特征是組成性活性激酶活性。一旦 Activin A 與其 II 型受體結(jié)合，兩個(gè) I 型受體就會(huì)被募集，并被 II 型受體磷酸化，這一事件導(dǎo)致其激 酶活性被激活。大多數(shù) TGF-β超家族成員與 ActRIIB 或 ActRIIA 結(jié)合，而只有少數(shù)配體與 ALK4 高親和力結(jié)合。除了典型受體激酶- smad 軸外，激活素 A 還被證明參 與 P38、ERK、PI3K、WNT 等途徑。


II. Description
